Extensively used standard buffer for extraction of high molecular weight DNA (>50-100 kb) from fungi and plants. Applicable for extractions of small scale up to large scale (ng up to g DNA amount).

Cetyltrimethyl ammonium bromide (CTAB), a cationic detergent, enhances cell disruption by denaturing and precipitating the cellwall lipopolysaccharides and proteins. In presence of Na+ concentrations above 0.5 M, the DNA will remain soluble. EDTA inhibits nuclease activity by binding divalent cations. Addition of PVP40000 has been proven to be useful for DNA extractions from polyphenol rich matrices (e.g. leaf material) - see CTAB Lysis Buffer + PVP40000.
